以pGEM-3Zf(+)为载体构建了痘苗病毒天坛株基因组HindⅢA、B、C片段约100kb共14个片段的无性繁殖系,并采用10种限制性内切酶就克隆的14个片段的主要酶切片段与WR株进行了比较。结果发现,在痘苗病毒天坛株基因组HindⅢ A片段的8个克隆片段中,有5个克隆片段的10种限制性内切酶的酶切片段与WR株相应克隆片段完全相同,其余3个克隆片段与WR株相应克隆片段相比,仅有1或2种限制性内切酶的酶切位点数不同;而痘苗病毒天坛株基因组HindⅢ B、C片段的6个克隆片段在大小和10种限制性内切酶的酶谱上与WR株相应克隆片段有很大差异。结合对过去已克隆的片段及国外已发表资料的分析,发现痘苗病毒天坛株与WR株基因组的主要差异表现在其基因组两侧区域即基因组HindⅢ B、C片段内。
Abstract:The Tiantan strain of vaccinia virus had been widely used for im munoprohylalaxis of smallpox in our country for many years. The already cloned Hind Ⅱ O-P fragments of this virus genome were mostly located on its central regions and no substantial differences were found between the genomes of Tiantan and WR strain. The Hind ⅡA, B, C fragments of the genome of vaccinia virus Tiantan were cloned by using pGEM-3Zf (+ ) as a vector, and all cloned fragments were compared with the corresponding fragments of WR strain genome by 10 restriction enzymes. The results showed that the Hindi A fragments of Tiantan and WR strain were basically identical, aside from the variability in three restriction enzyme sites; but the Hindi B, C fragments of Tiantan genome were distinctly different from those of WR genome in sizes and restriction maps. Therefore, according to our present and previous data, it is obvious that the most substantial variability between the Tiantan and WR strain genomes was located in the fragments of Hind Ⅱ B, C. The polymorphisms of the cloned fragments are also discussed.
