Abstract：ariations in the sequences of the Pre-core region of hepatitis B virus(HBV)genomes iso-lated from sera of chinese chronic hepatitis B patients were studied.The entire Pre -core re-gions of HBV DNA were amplified by polymerase chain reaction (PCR) from 8 anti-HBepositive and 2 HBeAg positive chronic carriers. The amplified fragment were either cloned intoM13mp19 bacteriophage vector and at least three clones from each sample were sequenced orsequenced by PCR-direct sequencing method.By comparing the Pre-core sequences to that ofa previously cloned chinese prototype HBV DNA sequece (pADR-I)10 types of Pre-coremutations were found Six out of 10 types have a TAG stop codon mutation at codon 28 ofthe pre-core region.Two types show mutation from G to A at nucleotide 1901.One typehas a mutation which eliminates the translational initiation codon of the Pre-core gene andanother one type has a point mutation of nucleotide 1824 from T to C which does not leadto amino acid alteration.Our data demonstrate that the most prevalent type of mutation inthe HBV Pre-core region among chinese patients is the generation of a translational stopcodon.
