Abstract:sing SMV─Sa RNA as template, SMV cDNA was synthesized by AMV reverse tran-scriptase. The synthesized cDNA was then amplified by polymerase chain reaction (PC.ADNA fragment with 0.8kb was obtained after 30 PCR amplification. The restriction map of the DNA fragment was analyzed. The results show the coat protein gene of SMV Sa strain has been cloned. The BamHI restriction endonuclease fragments of pSMV61 was labeled by nick translation and used in dot-blot hybridization, as little as 0.4ng of purified virus were de-tected. Spot hybridization can also be applied to detect SMV in soybean leaves.
