为鉴定此次安徽省境外输入新型冠状病毒的分子特征,本研究对两例病例进行了高通量测序和基因组序列分析。高通量测序获得两株病毒的基因组全长分别为29 858nt和29 858nt,利用NextStrain和2019新型冠状病毒信息库(https://ngdc.cncb.ac.cn/ncov/)在线分析对两例毒株全基因组核苷酸和氨基酸变异位点进行分析。将全基因组序列上传到(https://pangolin.cog-uk.io)和(https://clades.nextstrain.org/)网站,通过在线网站分析结果显示,两株病毒均为B.1.617.2变异株(Delta),NextStrain进化分析为21A。两株毒株分别发现了49和48个核苷酸突变位点,氨基酸的变异位点分别有35和34个,其中存在相同的7个同义突变。两株病毒主要在ORF7a基因上C27527T(P45L)的突变不同。这是安徽省首次发现境外输入的B.1.617.2变异株(Delta株)此变异株存在引发本地的流行和爆发的风险,需要严密持续的对境外输入病例进行流行病学调查和病毒基因分子特征分析。
Abstract:To identify the molecular characteristics of severe acute respiratory syndrome-coronavirus-2(SARSCoV-2)in Anhui Province,China. High-throughput sequencing was employed to obtain the full length of the genomes of viruses in two cases,and was found to be 29,858 nt and 29,858 nt. Analysis using the Internet databases NextStrain and 2019 Novel Coronavirus Database(https://ngdc. cncb. ac. cn/ncov/) revealed the nucleotide and amino-acid mutation sites in the entire genome of the two strains. We uploaded the whole genome sequence to two websites(https://pangolin.cog-uk.ioandhttps://clades.nextstrain.org),The online website analysis results showed that the two viruses were B.1.617.2 mutant strains(Delta),and NextStrain evolution analysis showed that they were classified as 21A. We identified 49 and 48 nucleotide mutation sites in the two strains. We discovered 35 and 34 amino-acid mutation sites,among which seven synonymous mutations were identified. The two strains differed mainly in the C27527T(P45L)mutation in ORF7a. This is the first time that an imported B.1.617.2 mutant(Delta strain)was found in Anhui Province. This mutant carries the risk of causing local epidemics and outbreaks. Epidemiological investigations and analysis of the genetic molecular characteristics of viruses imported from outside China must be carried out.
