采用细胞病变抑制法测定了病毒诱导前后草鱼外周血中干扰素的活性变化。结果表明,经病毒诱导的草鱼血清中出现明显的干扰素抗病毒活性,25℃水温下诱导3天,其活性达到高峰,在草鱼细胞中的效价(Log2CPEI50/0.1ml)可达11.08±0.58以上。草鱼血清干扰素具有100000g离心不沉降,耐热,在pH2.0-10.0范围内活性稳定,抗DNase和RNase,对胰蛋白酶、糖苷酶和NaIO4敏感等性质,其抗病毒作用依赖于细胞内RNA和蛋白质的合成。在鱼类细胞中具有抑制不同病毒的能力,但在人、哺乳类、鸟类和贝类等细胞中无抗病毒作用,表现出抗病毒的广谱性和相对的种属特异性,与高等脊椎动物干扰素的特性相一致
Abstract:The interferon activity was detected in sera of grass carp (Ctenocpharyngodon idellus) following infection with haemorrhagic virus by cell pathological effect inhibition assay. When the grass carp was induced at the temperature of 25℃ for 3 days, the serum samples provided the best interferon titres, its antiviral activity assayed on grass carp cells was higher than 11.08±0.58 (X±SD) log 2 units. Biochemical studies showed that the grass carp serum interferon was not sedimented at 100000g for 2 hours, exhibited stability to pH2-pH10 and heat (56℃), it was resistant to DNase and RNase, but was sensitive to trypsin, glucosidase and NaIO 4. Its inhibitory effect on virus replication was dependent on the process of cellular RNA and protein systhesis. It was able to inhibit different kinds of virus in the cells of grass carp, but failed in human, mammalian, avian and shellfish cells. The result showed that the grass carp serum interferon was specific to fish cells and viruses. Its biological characteristics were consistant with that of high vertebrate interferon.
