分析人间传染的高致病性病毒感染的动物模型应用情况,探讨人间传染的高致病性病毒在建立感染性动物模型中遇到的问题,为传染病防控科研和临床研究中动物模型选择和使用提供参考和依据。通过检索中国知网、万方数据库及PubMed数据库中人间传染的高致病性病毒感染动物模型的文献,对人间传染的高致病性病毒常用的感染性动物模型进行分析探讨。危害程度为第一类的高致病性病毒中,使用小鼠、猴及豚鼠为感染性动物模型的分别占95.83%、79.17%及62.50%;危害程度为第二类的高致病性病毒中,使用小鼠、猴及兔为感染性动物模型的分别占91.17%、55.88%及24.32%。使用实验小鼠作为感染性动物模型中,BALB/c小鼠和C57BL/6小鼠的比例分别占17.33%和13.33%。基因修饰小鼠归于其他小鼠类型,在其他类型占68.83%。使用实验猴作为感染性动物模型中,恒河猴和食蟹猴比例分别占37.18%和26.92%。相同科属的病毒使用的动物模型大致相同;其中还有16种高致病性病毒未检索到感染性动物模型的相关文章,包括类天花病毒、希普尔病毒、萨比亚病毒等。目前人间传染的高致病性病毒感染性动物模型使用最多的是小鼠,其次是猴;在使用的感染性小鼠动物模型中,BALB/c小鼠和C57BL/6小鼠的使用较多,基因修饰小鼠也被广泛使用;在使用的感染性猴动物模型中,食蟹猴及恒河猴使用最为广泛。对未检索到感染性动物模型的16种病毒,如类天花病毒、希普尔病毒、戈尔迪病毒等应提高关注,需探索建立以上病毒感染性动物模型的方法,为传染病的防控提前做好技术储备。
Abstract:This study analyzed the application of animal models for highly pathogenic human infectious viruses and discussed the challenges encountered in establishing infectious animal models. The paper aims to provide references for the use of animal models in infectious disease prevention and clinical research. The paper analyzed the published infectious animal models used in highly pathogenic human viruses through databases in CNKI, Wanfang, and PubMed. Among the infectious animal models used for the first-class highly pathogenic viruses published in the databases, the rate of use of mice, monkeys and guinea pigs was 95.83%, 79.17% and 62.50%, respectively. For the infectious animal models used in the second class of highly pathogenic viruses, the rate was 91.17% in mice, 55.88% in monkeys and 24.32% in rabbits. BALB/c and C57BL/6 mice accounted for 17.33% and 13.33% respectively when analyzing studies using infectious mouse models.Genetically modified mice accounted for 68.83% under other mouse types. In the analysis of studies using infectious monkey models, rhesus and cynomolgus monkeys accounted for 37.18% and 26.92%. Viruses within the same genus or family tended to use similar infectious animal models. In addition, we did not find relevant studies in infectious animal models for 16 highly pathogenic viruses, including Alastrim, Hepel and Sabia viruses. In conclusion, mice are the most commonly used infectious animal model in highly pathogenic human infectious viruses, followed by monkeys. Among the infectious mouse models published in the papers, the BALB/c and C57BL/6 strains are widely used, with genetically modified mice also playing an important role.Among the monkey models, the rhesus and cynomolgus monkeys are the most frequently used. More attention should be paid to the 16 viruses lacking established infectious animal models, such as Alastrim virus, Hypr virus, and Gordil viruses. Methods for establishing animal models for these viruses need to be explored in advance to prepare for effective infectious disease prevention and control.
