诺如病毒GⅡ.17新变异株引致福建省2014~2015年病毒性胃肠炎暴发

本文旨在鉴定福建省2014~2015年冬季导致病毒性胃肠炎暴发的病原体,并对病原体进行分子特征研究。收集福建省2014~2015年胃肠炎暴发期间急性病例的标本,开展诺如病毒检测并对衣壳蛋白基因片段进行分子特征分析。共检测3起暴发疫情标本31份,检出GⅡ型诺如病毒阳性标本17份。测序结果证实有13份标本为诺如病毒GⅡ.17新变异株,与2014年日本毒株Kawasaki 323同源性最高,在系统进化上形成一个新的独立的分枝,有别于近年来本地散发流行及全球暴发流行的优势基因型GⅡ.4。是福建省内首次检出诺如病毒GⅡ.17,并引起病毒性胃肠炎暴发。

Abstract:

We wished to delineate the molecular characteristics of the novel norovirus GⅡ.17 variant associated with an outbreak of acute viral gastroenteritis in Fujian Province,China,in the winter of 2014~2015.Noroviruses were detected in the specimens of patients suffering from acute gastroenteritis by reverse transcription-polymerase chain reaction(RT-PCR).The PCR products of positive samples were purified and sequenced.Partial sequencing of capsid genes was done using bioinformatics software and online databases,and a phylogenetic tree was constructed.Noroviruses were detected in 17 of 31stools.Analyses of 13 positive sequences indicated emergence of a novel norovirus GⅡ.17 variant,and this was confirmed to be the cause of the gastroenteritis outbreak.All strains shared a homology of 96.1%~97.3% with the Kawasaki 323 strain from Japan detected in 2014,and formed a new independent branch.These outbreak strains showed some differences from the local predominant strain,the 2012 Sydney GⅡ.4variant.We showed,for the first time,a norovirus GⅡ.17 variant that caused the viral gastroenteritis outbreak in Fujian in the winter of 2014~2015.More in-depth analyses of this norovirus GⅡ.17 variant could help to optimize preventative strategies and develop new and more effective therapeutic measures.
