Abstract:Foot-and-mouth disease virus(FMDV) is a highly contagious and acute infectious pathogen that seriously endangers the cloven-hoofed animals. With high mortality and robust transmissibility, the virus could traverses the geographic barriers to perpetuate its spread, rendering serious influence on healthy development of the livestock industry and international trade in animal-derived products. This typical pathogenic characteristic is closely associated with its unique genomic architecture, particularly the 5' untranslated region(5' UTR), a critical domain that ranks first among the family Picornaviridae in both length and functional complexity. The FMDV 5' UTR fulfils multiple regulatory functions throughout the viral life cycle, encompassing protein translation initiation, viral nucleic acid replication activation, immune signaling modulation, genomic RNA packaging and virion stabilisation, thereby serving as a critical structural element for the production of infectious viral particles in FMDV. Based on the viral replication cycle, this review summarizes the mechanisms of the FMDV 5' UTR in regulating viral proliferation, aiming to provide a theoretical reference for exploring viral biological characteristics and developing novel targeted antiviral therapeutics.
