EV-A71疫苗上市前后河北地区EV-A71流行株VP1区基因特征分析

摘要(Abstract):

了解河北地区EV-A71(Enterovirus A71)疫苗上市前后EV-A71遗传变异情况。在NCBI网站上获得疫苗株及EV-A71各亚型参考株序列。A亚型参考株为1969年加利福尼亚州分离株,NCBI编号为U22521。B1亚型参考株NCBI编号为AF135871;B2亚型参考株为AF009534;B3亚型参考株为AF376105;B4亚型参考株为AF376111;B5亚型参考株为AY258307;C1亚型参考株为AF376081;C2亚型参考株为AB213614;C3亚型参考株为AY125966;C4a亚型参考株为EU024958/AY905614;C4b亚型参考株为AY547499/AF302996;C5亚型参考株为EF063152;阜阳株为EU703813;疫苗株为HQ328793/EU812515/JX025561。对2011-2019年手足口病患者样本分离的EV-A71 VP1区核苷酸序列进行测定,对获得的序列进行遗传进化、相似性及位点变异等分析。测序成功的196条VP1区序列均属于EV-A71 C4a亚型,相同年份以及相近年份的分离株在进化树中存在聚集现象,各序列之间相似性较高。中和表位PEP27(site142-156)整体受到负向选择压力作用,其中VP1-145位点常出现VP1-145Q与VP1-145G突变;VP1区序列整体处于负向选择压力中。EV-A71疫苗上市前后河北地区EV-A71 VP1区遗传特征相对稳定,未出现较大的变异。
