自甲型肝炎患者粪便中分离到一株甲型肝炎病毒(TZ-84)。病毒不产生细胞病变,胞浆中有颗粒性荧光。细胞中的病毒抗原能被抗甲肝病毒抗体阳性血清所中和。 TZ-84株病毒耐酸,耐乙醚。免疫电镜见有大量直径27～30mm的甲肝病毒颗粒,以空心为主。将其接种细胞后的黑暗期随传代而缩短,病毒滴度则随传代而增加。将细胞粉碎后获得的病毒。ELISA滴度为1:16。用其检测45份人血清中抗HAV-IgM抗体,并与Abbott ELISA药盒比较,符合率为91.1％,敏感度为87.1％。

This paper reported that a strain of Hepatitis A virus was isolated and propagated in human diploid cell line(2BS).It was isolated from the feces of a patient with early HAV infection and named TZ-84. Evidence of viral multiplication was obtained by demontration the immunofluoresence, ELISA, and the visualization of 27-30nm particles in cell extracts by IEM. Virus was propagated more successfully in 2BS than in Vero.No cytopathology was observed and no detectable amount of HAV was present in the tissue culture supernatant throughout the study period.The specificity of TZ-84 strain of HAV was demontrated by serum neutralization rea'ction in tissue culture, ELISA blocking assay, and immunofluore-scent test with McAb to HAV.TZ-84 was stable to ether and acid (pH3.0) . All of these tests show that TZ-84 is a strain of HAV.TZ-84 strain HAV has been successfully passaged in cell culture.The eclipse period in 2BS was shortened with passage and the amount of HAAg in cell culture was increased. The ELISA titer of HAAg in cell culture extracts was 1:16 after 10th passages.45 sera were tested for anti-HAV IgM with TZ-84 and the results compai-red with Abbott ELISA Kit. The coincidence rate was 91. 1% and the sensitivity was 87.1%.TZ-84 strain of HAV can be used for production of HAAg and for diagnostic use and possibly for vaccine study.
