流行性出血病病毒(Epizootic haemorrhagic disease virus,EHDV)是一种严重危害反刍动物的虫媒病毒,全世界已发现9种血清型EHDV,长久以来我国EHDV血清型分布、病毒活动规律和病毒遗传特征不明确,对我国的畜牧业进出口和牛羊养殖业构成了潜在威胁。本文首次报道了2013~2016年在云南省与广西壮族自治区设立的哨兵动物中,9株EHDV-5型毒株的分离以及病毒基因节段2、3与6(Seg-2、Seg-3、Seg-6)的序列特征。中国分离的9株EHDV-5型毒株Seg-2,Seg-3与Seg-6及其编码蛋白之间高度保守,核酸与氨基酸序列相似度均大于99%,在系统发生树上聚为紧密的一簇,表明我国云南与广西流行的EHDV-5有着最近的共有祖先。中国与澳大利亚EHDV-5毒株不同基因节段之间也有着较高的序列相似度(>91%),表明两地流行EHDV-5型毒株有着共同的起源。中国毒株的Seg-2与Seg-6在系统发生树上虽与澳大利亚毒株(CSIRO 157)聚为一簇,但最终形成了独立的进化分支,表明中国流行的EHDV-5发生了独立的进化。本研究首先丰富了世界范围内EHDV-5型毒株资源,其次为我国开展EHDV-5病毒的更深入研究提供了宝贵材料和理论数据。
Abstract:The epizootic haemorrhagic disease virus(EHDV)is an arbovirus which is harmful to ruminants.There are nine serotypes of EHDV worldwide,but the epidemiology and hereditary characteristics of the EHDV in China are still unclear,which could seriously endanger the cow-breeding industry. We report nine strains of EHDV-5 and the sequence characteristics of the virus gene segment 2(Seg-2),Seg-3 and Seg-6 isolated from sentinel animals in Yunnan Province,China,and Guangxi Zhuang Autonomous Region,China,from 2013 to 2016. Sequencing of Seg-2,-3 and-6 and coding proteins showed highly conserved regions among the nine strains of EHDV-5. The similarity between the nucleotide sequence and amino-acid sequences were >99%. A close cluster in phylogenetic trees indicated that the nine strains of EHDV-5 had a common ancestor.There was also high sequence identity(>91%)between the different gene segments of the EHDV-5 strain in China and Australia,which indicated that the strains had a common origin. The Seg-2 and Seg-6 of Chinese strains clustered with Australian strains in the phylogenetic tree,so they eventually formed an independent evolutionary branch,indicating that EHDV-5 isolated from China evolved independently. This study enriches the resources of EHDV-5 strains worldwide and lays a foundation for the study of the genetic characteristics,pathogenicity and etiological diagnosis of the EHDV-5 in China.
